In the past decade, Supercritical Fluid Chromatography (SFC) has increasingly gained momentum in both chemical and pharmaceutical industries. The intrinsic low viscosity and high diffusivity of supercritical CO2 has rendered SFC higher separation speeds and efficiency than traditional LC, especially in chiral analysis and purifications. As the analytical philosophy in drug discovery gradually shifts towards generating high-quality and more informative data within a minimal time frame, SFC MS readily lends itself as an attractive complement to the industry-dominating LC MS, due to the combination of the high speed and unique selectivity of SFC, and the universality, sensitivity, and specificity of MS.

In this webinar, Dr Andreas Tei reviews the history and the fundamentals of SFC-MS, from the perspectives of ionization and SFC hardware. SFC's orthogonality to LC is demonstrated by a wide variety of real industrial applications, including structure elucidation, degradant characterization, metabolite identification, pharmacokinetic measurement, and mass-directed purification. In addition, the comparison between LC-MS and SFC-MS in pharmaceutical applications and some recent endeavors in employing SFC-MS for high throughput analysis and method development are also introduced.
